Our Lady of Guadalupe Church is about two miles west of San Antonio's downtown River Walk area. The parish ministers to a low-income community which is mostly Mexican-American. Liturgies are rich in Native American and Hispanic culture.

The Jesuit Fathers of the New Orleans Province have been serving in the parish since 1932. Three, priests and one brother live and work in the parish.

The church is the Regional Sanctury of Our Lady of Guadalupe. As such it attracts many visitors and persons whose faith in the Protectress of the Americas leads them to pray at this shrine. The annual celebration of Our Lady's feast on December 12th is a rich and inspiring occasion. The dancing of Matachines and the singing of Mariachi begin at twilight on December 11th and continue for the following 24 to 30 hours.

Because of the number of persons who come to pray, the church is opened around 7:00 am and remains open until 5:00 pm each day of the week.

The church faces the Plaza Guadalupe on the south. Pope John Paul II celebrated Mass in this Plaza on September 13, 1987 during his visit to San Antonio.
